Unread is what every inbox shows: a message arrived, nobody opened it.
Unreplied is the one that saves sales: the customer sent the last message and nobody has answered, even if someone opened it. Reactions and read receipts do not clear it; only a real reply does. Sort by unreplied and you have the “customers waiting on us right now” list.

Do I need the WhatsApp Business API for this?
For a shared team inbox, yes: the API is what lets more than one phone see the same chats. Any provider works; you add your provider's credentials in the app.
What is the difference between unread and unreplied?
Unread means a message arrived and nobody opened it. Unreplied means it was opened but the last message is still the customer's. Both are shown; only a real reply clears unreplied.
Can callers see chats for leads that are not theirs?
That is a permission. By default a caller sees their own leads' chats; the owner sees all.
